Study of Recovery Rates of Motility of Spermatozoa in Cryopreservation

Abstract
Cryopreservation of spermatozoa is one of the good methods of treatment of infertile couples due to male factor. The objectives of this study were to compare the effectiveness of three cryoprotectants and to compare the effectiveness of postthaw
recovery rate of spermatozoa motility among various combination methods of rapid and slow freezing and thawing on each cryoprotectants. Semens of 46 healthy patients and donors were used. Cryoprotectants were 10% glycerol, GEYC (Glycerol-egg
yolk-citrate; Ackerman's cryoprotectant medium) and modified GEYC. The rapid freezing, slow freezing, rapid thawing and slow thawing semen were performed by programmed protocol. Data were evaluated by one-way ANOVA, two-way ANOVA and ANCOVA. @ES

results of this study were summarized as follows:
@EN 1. The best cryoprotectant was GEYC among three cyoprotectants.
2. Generally the slow freezing method in cryopreservation has the better recovery rate of spermatozoa using slow thawing method than rapid thawing method. The rapid freezing method in cryopreservation has the better recovery rate of spermatozoa
using
rapid thawing method than slow thawing method.
3. The best recovery rate was obtained from the method of slow freezing and slow thawing using GEYC cryoprotectant. Therefore the selection of the cryoprotectant and the combination of method of freezing and thawing must be taken into
consideration
in
cryopreservation of spermatozoa.
